Gene Discovery

Based on the phylogenetic relationships within the genus Meloidogyne, and the biological differences between species (such as host range, susceptibility of host R-loci, etc.) we have chosen 6 species for EST sequencing. As of August 2001, we have processed (trimming, Phred20, etc.) annotated and entered into GenBank dbEST: M. incognita: 5,713 ESTs, M. javanica: 3,994 ESTs, M. arenaria: 568 ESTs

The NemaGene clustering tool has revealed more than 2,000 Meloidogyne genes from our initial sequencing, which likely accounts for >10% of the gene complement. Annotation of the M. incognita v1.0 clusters showed that 76% of identified transcripts have significant database matches in other organisms (BLAST, E<10-5). Matches to other species were dominated by C. elegans; 66% of transcripts matched a C. elegans gene, and 52% had a C. elegans gene as their best matching sequence.
