Confirmation of staining specificity: Recovery and analysis of the histological stain.The signal detected as stain in tissue sections, which corresponds to the reverse-transcribed RNA, is DIG-labeled PCR product which can be recovered and analyzed, including by sequencing. An anticipated 230 bp, DIG -labeled product is identified in supernatant retrieved from rhizobia induced nodules in Medicago, stained for Mt-phan (Mt-phan). No product was detected in supernatant retrieved from no Taq control of Mt-phan stain in nodule sections (Mt-phan no T), and in that retrieved from no primer control of 18S rRNA staining in meristem section (18S no P).
