Identification and characterization of tDNA insertion mutants of Arabidopsis PI 4-kinase

The Wisconsin Biotechnology Center (Madison, WI) offers pools of Arabidopsis seeds which carry random tDNA insertion mutations. The seeds can be screened for tDNA insertions in a particular gene by PCR, using primers specific for the gene of interest and the tDNA flanking regions. We have identified a pool of seeds containing a tDNA insertion within a PI 4-kinase gene and are currently in the process of isolating the particular seed. The phenotypical and biochemical analysis of the mutant plants may help delineate cellular functions of PI 4-kinase and phosphoinositides.
