Identification
and characterization of tDNA insertion mutants of Arabidopsis

The Wisconsin Biotechnology Center (Madison, WI) offers pools of Arabidopsis
seeds which carry random tDNA insertion mutations. The seeds can be
screened for tDNA insertions in a particular gene by PCR, using primers
specific for the gene of interest and the tDNA flanking regions. We
have identified a pool of seeds containing a tDNA insertion within
a PI 4-kinase gene and are currently in the process of isolating the
particular seed. The phenotypical and biochemical analysis of the
mutant plants may help delineate cellular functions of PI 4-kinase
and phosphoinositides. |
